To determine the local resistance that a migrating cell is experiencing, we aim to develop computer models with cellular resolution that predict emerging tissue mechanical properties and the apparent tissue stiffness experienced by a migrating cell. The geometry of the cellular assemblies in the model will be taken from 3D microscopy images of tissues labeled with a membrane fluorescent marker (similar to Retzer K., Akhmanova M., et al. Nat Comm, 2019) thus the in silico tissue will mimic the cellular in vivo shapes. Using the genetic toolbox in Drosophila to induce mechanical perturbations, and by measuring local mechanical properties with a variety of available methods we will validate the model predictions.
